Before You Start

You’ll need:

  • MycoNutrients® sterile bag(s): MycoOne™ (All‑in‑One), or MycoSpawn™ (sterile grain) + MycoCoir™/MycoWood™ substrate
  • Inoculant: sterile liquid culture (LC) or uncontaminated spore syringe (purchased separately)
  • 70% isopropyl alcohol (spray), lighter/alcohol lamp, paper towel, clean gloves, marker pen
  • Laminar flow hood or Still Air Box (recommended). If not available: small room, doors/windows closed, no fans

Tip: Starting with spores? Consider converting to LC first for faster, cleaner colonisation.

Universal Sterile Workflow (Steps 1–10)

Follow these steps for every product, then continue to the correct section.

  1. Prepare your space – wipe the work surface with 70% Isylpropyl Alcohol (IPA); let dry. Close doors/windows; no airflow.
  2. Prepare yourself – wash hands; put on clean gloves; wipe with IPA (optional: mask/lab coat).
  3. Stage your items – bag, inoculant, needle, lighter, wipes, marker within easy reach.
  4. Sanitise contact points – wipe the self‑healing injection port and the area around it.
  5. Flame‑sterilise the needle – heat to red/orange; cool 10–15 sec (you can briefly touch to a fresh IPA‑damp towel after cooling).
  6. Inoculate – inject spores or LC through the self‑healing port.
  7. No sealing required – the self‑healing port reseals automatically. Lightly spritz the area if you wish.
  8. Label (recommended) – strain/culture, date, bag type/weight.
  9. Incubate – 20–24 °C, upright, indirect light; avoid direct sun and compressing the filter area.
  10. Check for growth – LC: ~3–5 days; spores: ~7–14+ days. If growth stalls, gently break & shake to redistribute.

All-in-One Bags (MycoOne™)

When to start fruiting: Wait until the bag is 100% white and firm, then allow an extra 72 hours to consolidate.

Choose your opening method (both work):

  1. “X‑cuts” (simpler, higher humidity): Using sanitised scissors, cut 2–4 small X‑shaped slits (2–3 cm) on the faces of the block where you want mushrooms to form.
  2. “Lid/Tent” (more fresh air): Cut the bag across the top and down two sides (leave one long side attached), then fold the plastic back like a lid to tent over the block. Open more for extra fresh air; close further if drying.

Fruiting conditions (typical):

  • Temperature: 18–22 °C
  • Humidity: 85–95% RH (aim for light condensation on bag walls)
  • Fresh air: Briefly fan or crack tent 1–2×/day, or use the lid/tent method for passive FAE
  • Light: Indirect/bright room light (e.g., 12/12 cycle); no direct sun

How to mist:

  • Mist the inside bag walls 1–2×/day with 2–4 light sprays. You want beads on the plastic, not water pooling on the block.
  • Do not soak pins/caps. If the block looks dry, mist above it so micro‑droplets settle gently.
  • If walls already show steady condensation, skip misting to avoid over‑saturation.

Reading your mushrooms:

  • Long stems, tiny caps → increase fresh air (open tent more / add an extra small X‑cut).
  • Cracked/dry caps → increase humidity (mist walls more / reduce opening size).

Harvest: Twist or cut clusters at the base when caps are mature. Remove leftover stumps to keep the surface tidy.

Multiple flushes (rehydration & rest):

  1. Tidy: After harvest, remove stumps/debris.
  2. Rehydrate (bottom‑water): Pour bottled mineral water into the bag so it pools around the block base. Guide:

    ~0.9 kg bag: 100–150 mL
    ~1.9 kg bag: 200–300 mL
    ~4.5 kg bag: 400–600 mL

    Let absorb for 6–12 hours, then pour off any excess.
  3. Rest the block: Close the bag/tent and keep humidity high for 2–5 days with minimal disturbance.
  4. Resume fruiting: Re‑open using your chosen method; maintain humidity & fresh air. Expect 2–3 flushes total in good conditions.

Side‑pinning? It’s normal. Keep edges shaded, and avoid big air gaps down the sides when forming the block.

Grain + Substrate (MycoSpawn™ + MycoCoir™ / MycoWood™)

Step 1: Colonise MycoSpawn grain — follow the Universal Workflow. LC usually shows growth in 3–5 days, spores 7–14+. Break & shake when bag is at ~30–50% colonisation (white and firmness) to spread mycelium.

Step 2: Prepare to mix

  • Sanitise your work surface again; wipe scissors/knife with 70% Isopropyl Alcohol (IPA).
  • Have your sterile substrate ready (MycoCoir™ for CVG strains; MycoWood™ for wood-lovers).

Step 3: Mix (ratio) — open the substrate bag in front of a laminar flow hood or in a still air box; add the fully colonised grain. Aim for ~1:6 to 1:8 spawn:substrate by weight. Reseal the bag with a bag sealer ensuring you get some fresh air from the laminar flow hood in the bag. Mix quickly and evenly; minimise open time. If using a monotub: close the tub and seal the holes with micropore tape.

Step 4: Colonising the substrate bag — 7–14 days at 20–24 °C until the substrate is fully white and firm.

Step 5: Prepare for fruiting — choose your opening method:

  1. “X-cuts” (simpler, higher humidity): Using sanitised scissors, cut 2–4 small X-shaped slits (2–3 cm) on the bag faces where you want mushrooms to form. Works well if your room is dry, as it helps retain humidity.
  2. “Lid/Tent” (more fresh air): Cut the bag across the top and down two sides (leave one long side attached). Fold the plastic back like a lid to tent over the block. This increases fresh air exchange (FAE) and improves cap quality. If the block dries, close the lid further or mist more often.
  3. Monotub method: If you used a monotub, remove tape/adjust vent covers for passive FAE. Maintain high humidity by misting tub walls.

Step 6: Fruiting conditions:

  • Temperature: 18–22 °C
  • Humidity: 85–95% RH (aim for light condensation on bag/tub walls)
  • Fresh air: Briefly fan or crack lid once or twice daily, or adjust tent/vents for passive exchange
  • Light: Indirect/bright room light (12/12 cycle is ideal); no direct sun

Step 7: How to mist:

  • Mist the inside bag walls or tub walls 1–2×/day with 2–4 light sprays.
  • You want beads of water on the walls, not pooling on the block surface.
  • Do not spray pins/caps directly — if block looks dry, mist above it so droplets settle gently.
  • If condensation is already steady, skip misting to avoid over-saturation.

Step 8: Reading your mushrooms:

  • Long stems, tiny caps → increase fresh air (open tent more / adjust tub vents).
  • Cracked/dry caps → increase humidity (mist walls more / reduce opening size).

Step 9: Harvest: Twist or cut clusters at the base when caps are mature. Remove stumps and debris to keep the surface clean.

  3. Rest the block: Close the bag/tub and maintain humidity for 2–5 days with minimal disturbance.
  4. Resume fruiting: Re-open with your chosen method; maintain humidity & FAE. Expect 2–3 flushes total in good conditions.

Troubleshooting

  • No growth by Day 10 — LC: re‑check temp (20–24 °C) and do a gentle shake. Spores can take 14+ days. If still nothing by Day 14–21: suspect dead/contaminated inoculant; restart with a trusted source.
  • Sour smell / yellow slime — likely bacterial; discard safely and review sterile steps.
  • Green/blue patches (not bruising) — likely mold (Trichoderma); discard safely.
  • Stalled at 70–90% — break/shake (grain) or remix top layer (substrate). If no progress in 7–10 days, restart.
  • Pins won’t form — increase fresh air slightly, ensure indirect light, maintain 85–95% RH (mist walls, not block).
  • Leggy stems/tiny caps — try the lid/tent method (see All‑in‑One) for more fresh air exchange (FAE).
  • New to sterile work? Contamination risk is higher with separate grain + substrate without a hood/SAB. Consider starting with MycoOne™ All‑in‑One.
